Site-specific click chemistry-mediated scaffold engineering to bioorthogonally functionalize cell surface in cell-based therapeutics

Engineering of bioorthogonal-mediated chemical scaffolds and/or tags to develop cell therapeutics requires a rational design approach. This challenge in cell therapeutics can be addressed by developing promising scaffolds and/or tags that function under specific physiological conditions. Target interactions and efficiency can be precisely regulated within defined cellular environments by incorporating highly reactive, reaction-specific entities into cell-based therapeutics. This enables on-target activation, enhanced stability, biocompatibility, and improved performance under tumor-specific conditions. This review primarily focuses on: (1) the importance of bioorthogonal-based scaffolds, (2) selected bioorthogonal reactions and their associated kinetics, (3) the use of these scaffolds for biomolecular engineering, and (4) the role of scaffolds and/or tags in the development of cell-based therapeutics. Various bioorthogonal chemical tags, together with their reactions and kinetics, are systematically highlighted. The use of these scaffolds to develop therapeutics, cell-cell interactions, and/or cellbased therapeutics is also discussed.
