Ex vivo priming offers an opportunity to enhance NK cell antitumor activity prior to therapeutic application. Herein, NKG2D-targeting peptide-nanoparticle conjugates were developed as a clustering-driven priming platform. Multivalent presentation of NKG2D-binding peptides induced NK cell clustering, enhanced cytokine secretion, and improved cytotoxicity against MDA-MB-231 cells.
